The response of four winter wheat cultivars, differing in resistance to Fusarium head blight (FHB), to spray inoculation with four selected Fusarium graminearum isolates, mixture of these isolates and frequently used F. culmorum isolate B was studied in five field and glasshouse experiments during 2008-2010. Analyses of variance showed highly significant main and interaction effects of cultivar, inoculum source and environment (year-trial) on all five examined traits indicative of disease severity, yield loss and accumulation of mycotoxins. The relations between traits were not evidently influenced by the used isolate. Resistance...

In an attempt to speed up the process of screening of transgenic cotton (G. hirsutum L.) plants, a visual and rapid loop-mediated isothermal amplification (LAMP) assay was adopted. Genomic DNA was extracted from fresh leaf tissues of T2 transgenic cotton containing chitinase (chi) and cry1A(b) genes. Detection of genes of interest was performed by polymerase chain reaction (PCR), LAMP and real-time PCR methods. In LAMP assay the amplification was performed after 30 min at 65°C when loop primers were involved in the reaction. The involvement of loop primers decreased the time needed for amplification. By testing...

Yellow mustard, cultivated in northern Shaanxi of China, is a valuable germplasm of Brassica juncea with low erucic acid content. Its yellow seed colour is controlled by a recessive allele of a single gene, whose dominant allele conditions brown seed colour. To map the yellow seed colour allele, amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR) technologies were used to identify markers linked to the recessive allele. The analysis was done on 386 F2 plants, segregating for seed colour, from the cross Wuqi yellow mustard × Wugong mustard. The plants were selfed to determine their seed colour genotype....

The presence of genes for resistance to scab (Venturia inaequalis) and powdery mildew (Podosphaera leucotricha) was studied using molecular markers in a sample of 279 apple cultivars from the Czech collection of apple genetic resources. The sample comprised 37 cultivars supposed to have the Vf gene for scab resistance, 97 reference world cultivars and 145 old and local cultivars. Six PCR molecular markers for the scab resistance genes Vf, Vm, Vbj, Vr and Vh and three PCR molecular markers for the powdery mildew resistance genes Pl-w, Pl-1 and Pl-d were...

The prolamin (avenin) patterns of oat (Avena sativa L.) cultivars released in the Czech Republic and in the former Czechoslovak Republic were analysed by acid polyacrylamide gel electrophoresis (A-PAGE). Forty-nine oat (Avena sativa L.) accessions of domestic origin, maintained in the Czech collection of oat genetic resources, were analysed. The evaluated set contained 18 modern and 31 old cultivars. Thirty accessions showed a homogeneous prolamin pattern. The other accessions were heterogeneous with two or three different patterns, present at different percentages. Heterogeneity was present in 48% of the old cultivars, but ...
