Genealogical Analysis in the Czech Spring Wheat Collection and its Use for the Creation of Core Collection

A spring wheat collection of the Czech gene bank included altogether 3270 original accessions in 2005; among them 2123 accessions with pedigree were analyzed using the construction of genetic profiles and subsequent calculation of Renkonen similarity indexes. Genetic diversity was estimated by Shannon diversity index. Subsequently we calculated cluster analysis using coefficients of parentage and in addition, we selected representatives of particular clusters for a core collection. Physiologic Specialization of Wheat Leaf Rust (Puccinia triticina Eriks.) in theCzech Republic in 2001-2004

In 2001-2004 virulence of the wheat leaf rust population in the Czech Republic was studied on Thatcher near-isogenic lines with Lr1, Lr2a, Lr2b, Lr2c, Lr3a, Lr9, Lr10, Lr11, Lr15, Lr17, Lr19, Lr21, Lr23, Lr24, Lr26 and Lr28. Samples of leaf rust (180 in total) were obtained from different parts of the Czech Republic. Resistance genes Lr9 and Lr19 were effective to all tested isolates like in the previous years. Estimation of Fatty Acid Content in Intact Seeds of Oilseed Rape (Brassica napus L.) Lines Using Near-Infrared Spectroscopy

Based on NIRS (near infrared reflectance spectroscopy) measurements carried out in a collection of 262 samples of winter oilseed rape with a different content of fatty acids (FA) in oil, calibration equations for the laboratory instrument Foss-NIRSystem 6500 were developed. Progression of Deoxynivalenol Concentrations in Spikes and Kernels of Winter Wheat Cultivars after Inoculation with Fusarium culmorum

Progression of deoxynivalenol (DON) concentrations in spikes and kernels was studied in relation to Fusarium head blight (FHB) symptoms in five winter wheat cultivars, differing in resistance to FHB, after single floret inoculation with an aggressive isolate of Fusarium culmorum. After inoculation in field conditions the spikes were detached from the plant and kept in the greenhouse under controlled conditions. The modified PAGE method (TRIS - Glycine buffer pH 8.9) was used for the characterisation of selected 25 registered potato varieties. This method enabled to identify each variety from the examined variety set. The calculation of identity indexes (proportion of common bands) helped to evaluate the similarity of varieties from these aspects. The examination of electrophoretic profiles of soluble tuber proteins, which are highly polymorphic and stable, can be considered as valuable for variety characterization and identification.
