Achievements and prospects of wheat breeding for disease resistance in the world and in theCzech Republic are discussed. Attention was paid to possibilities of increasing resistance to rusts, powdery mildew, Fusarium head blight, leaf blotch, glume blotch, tan spot, common bunt and barley yellow dwarf virus on wheat. Methodical approaches adopted in national ring infection tests were outlined. New sources of resistance to the above-mentioned diseases were detected and described on the basis of three-year results of field infection tests.

The Barley Microarray. A Community Vision and Application to Abiotic Stress

A microarray chip representing approximately 20 000 barley unigenes was produced as part of a USA project entitled "An integrated physical and expression map of barley for Triticeae improvement". The content of the chip was derived from more than 400 000 barley "Expressed Sequence Tag" (EST) sequences received from cooperators inUSA,Germany,Australia,Japan,Scotland, andFinland, plus about 1000 sequences retrieved from the GenBank nr database or GrainGenes. Genetic and Physical Mapping of Genic Microsatellites in Barley (Hordeum vulgare L.)

Due to the availability of sequence data from large-scale EST (expressed sequence tag) projects, it has become feasible to develop microsatellite or simple sequence repeat (SSR) markers from genes. A set of 111 090 barley ESTs (corresponding to 55.9 Mb of sequence) was employed for the identification of microsatellites with the help of a PERL5 script called MISA. As a result, a total of 9 564 microsatellites were identified in 8 766 ESTs (SSR-ESTs). Cluster analysis revealed the presence of 2 823 non-redundant SSR-ESTs in this set. Powdery Mildew Resistance of Czech and Slovak Spring Barley Breeding Lines in Variety Trials

In 2001-2005, resistance to powdery mildew was studied in 227 Czech and Slovak breeding lines of spring barley included in the breeding station trials or official trials. Seventeen known resistances were identified (Al, Ar, At, HH, Kr, La, Ly, Mlo, N81, Ri, Ru, Sp, St, Tu, We, Mla21, and Mlp1). Unknown resistances were found in 11 breeding lines, in five of which resistance was effective against all used pathotypes of the pathogen. Oligogenic Inheritance of Resistance to Plum Pox Virus in Apricots

In order to determine the inheritance of resistance to PPV in apricot three crosses between resistant and susceptible cultivars and selections were performed. The B1 seedlings were inoculated with the PPV-M strain by an infected bud. PPV infection was evaluated over 5 consecutive growth periods through visual symptoms, ELISA and in some cases reverse transcriptase PCR assays. Chi-square analysis of each B1 progeny was performed to determine if the segregation ratio differed from the expected ratio.
